The effector/target ratio represents the ratio of NK cells to cancer cells. Percent cytotoxicity - the percentage of cancer cells that were destroyed in the cancer cells analyzed - varied with different effector/target ratios. Effector/target ratios of 10 and 5 indicated that NMN had significantly enhanced cytotoxicity in the liver, while an effector/target ratio of 100 was optimal for NMN efficacy in the spleen. Using the same measure of cytotoxicity, the team then sought to determine whether oral NMN had similar benefits. Oral supplementation of NMN at 625 mg/kg significantly enhanced NK cell cytotoxicity in Takeda and Okumura's 32-week-old mice for analysis. Neither injection nor oral administration of NMN increased NK cell numbers . This result strongly supports that NMN supplementation improves overall NK cell activity in mice, as NK cells show improved, potent cancer cell destruction capacity without becoming more immune cells .
Optimal effector/target ratios - the ratio of NK cells to cancer cells - were 5, 10 and 20 for the liver and 200 for the spleen. At these ratios, the percentage of destroyed cancer cells, the percentage of cytotoxicity, was significantly increased at the 625 mg/kg/day dose (solid squares in the figure).

What is the role of NAD+ in fat accumulation?
Adipogenesis -- the maturation of fat cells -- is a finely controlled process. These cells derive from precursors called preadipocytes, a process regulated by the fine control of gene regulatory proteins called transcription factors that coordinate the expression of hundreds of proteins. When preadipocyte differentiation is disrupted, adipocytes swell and ectopic lipid accumulation occurs in inflammation and metabolic dysfunction.
Metabolic changes accompany lipogenesis. Although several studies have been performed to determine the metabolic changes that occur during preadipocyte differentiation, little attention has been paid to the role of NAD+ metabolism.
Upregulation of NAD+ synthesis is required for preadipocyte differentiation
Okabe and colleagues sought to elucidate the metabolic changes that occur during preadipocyte differentiation. They found that levels of NAD+ and NMN, a NAD+ precursor, increased significantly during preadipocyte differentiation. Okabe and colleagues next examined whether the adipogenesis program is affected by NAD+ levels. Alpha-ketoglutarate regulates PPARγ activation in preadipocyte differentiation
To activate a gene, the PPARγ DNA sequence must be demethylated, removing molecular tags called methyl groups that bind to DNA. The researchers found that when demethylation was inhibited, levels of PPARγ activation and preadipocyte differentiation were inhibited.
In addition, the metabolite α-ketoglutarate is a known cofactor for demethylases. Interestingly, levels of α-ketoglutarate were significantly increased during preadipocyte differentiation, which correlated with increased levels of NAD+. The researchers then wanted to see if there was a link between NAD+, alpha-ketoglutarate, and lipogenesis. To do this, they supplemented the preadipocytes with alpha-ketoglutarate. This rescued PPARγ levels and preadipocyte differentiation, all of which were inhibited by inhibiting NAD+ synthesis. Alpha-ketoglutarate prevents diet-induced obesity by promoting proper lipogenesis
Finally, Okabe and colleagues investigated how NAD+ and alpha-ketoglutarate affect obesity. Mice fed an obesity-inducing high-fat, high-sucrose diet were orally administered alpha-ketoglutarate in drinking water. Surprisingly, alpha-ketoglutarate administration completely suppressed diet-induced obesity, similar to mice fed a normal diet.
The researchers also examined the physical characteristics of the mice's fat tissue. All nine patients had fever, cough and lethargy, which made them mostly bedridden. Loss of smell (anosmia) in 6 of 9 patients and diarrhea in 5 of 9 patients.
The patients were experiencing symptoms of COVID-19 disease when Huizenga started giving them the NMN cocktail, starting the dosing within 5 to 34 days of the onset of COVID-19 symptoms. Four of the nine patients were critically ill with deteriorating oxygen saturation levels, but the NMN cocktail caused rapid clinical improvement, including fever reduction within two to three days. Importantly, no patients required intensive care after treatment.
